Th17 cells play key roles in immune defense and tissue homeostasis but can also become pathogenic and induce autoimmunity. Gaublomme et al. (pp. 1400–1412) use single-cell genomics to reveal the cellular circuit that regulates Th17 cell pathogenicity, and Wang et al. (pp. 1413–1427) show that CD5L acts as a molecular switch that alters lipid metabolism to regulate Th17 function.
